If you can see the attached screenshot, if I want to store the backup in secondary destination, will the backup run on both the primary and secondary destination simultaneously on the scheduled time?

You don‘t need to configure a second backup job for the second location.
Use Backup copy Job instead of a backup job.

No, the backup will be produced first in the primary destination (backup repository) as selected in your screenshot. It will then be copied to the secondary destination afterwards depending on type of backup copy job for example.

P.S. if your primary destination is a scale-out backup repository and you want your secondary backup copy to go to object storage you will not need a backup copy job and instead you can leverage the capacity tier functionality: https://helpcenter.veeam.com/docs/backup/vsphere/capacity_tier_copy.html?ver=110
Guys, you are talking about backup copy and that is not the question. I am talking about backup job.
While configuring a backup job, when I set the primary repo and if I want to store in another repo which is by enabling “configure secondary destination for backup job” after configuring, will the backup job run simultaneously on the both repos based on the schedule?

Backup Copy Jobs always runs after the backup is done. There are two modes to choose:
